Leave the default settings for a full backup and click 'OK' to begin the backup process.

With the full backup complete, next the software automatically searches for any available firmware updates. In my case, a firmware update was available. I'll click 'Yes' to download and install the firmware update now.

Click 'Yes' to confirm that you want to install the new firmware.
*Important Note: be sure that your battery is fully charged before installing any updates. If you lose power during an update, the GPS won't work and you'll have to restore from the backup you made earlier.

The update downloads to your PC first, and then is installed onto the GPS (shown below). Wait for the update to finish downloading...

After downloading the update, Content Manager transfers the updated firmware to the GPS.

Click 'OK' after the firmware update completes.
Magellan's software doesn't tell you what to do next (the update program just unceremoniously exits after you click 'OK'), but you'll want to disconnect the GPS from the computer next and let the new firmware finish installing. That can be a mildly scary process, during which your Magellan Maestro will reboot several times, the screen will go blank, and some cryptic messages will appear on the screen. Don't worry, this is normal.
After the firmware finishes installing on your GPS, and the Main Menu appears on the device, again connect the Maestro to the PC via USB.

After connecting the Maestro to the PC, launch the Content Manager program (if it isn't already running).

After Content Manager loads, click on the blue update icon in the upper left portion of the screen.

Magellan's software doesn't provide any details about the available update, other than that it's 262 MB in size. Click 'Yes' to begin downloading.

Depending on your internet connection speed, the download will take anywhere from a few minutes to an hour.

Up-to-date
After the update downloads and transfers onto the Maestro 4350, Content Manager reports that "All files are up-to-date", which is a complex way of saying that your Maestro is up to date.
Disconnect the GPS from the computer to allow the update to finish installing. Your Magellan Maestro 4350 is now up-to-date.
